Target Information
Mouse phosphofructokinase, platelet isoform (PFKP) is a 784-residue cytosolic tetramer that catalyzes the ATP-dependent phosphorylation of fructose-6-phosphate to fructose-1,6-bisphosphate. Its N-terminal catalytic domain contains the conserved 244-Gly-Gly-Ala-Ser-Asn-248 motif, while a C-terminal regulatory region binds ATP, citrate and ADP to control glycolytic flux. Constitutively expressed in megakaryocytes, spleen and brain, PFKP localizes to cytoplasm and cytoskeletal junctions. Antibodies raised against the catalytic or regulatory peptide detect native, phosphorylated and acetylated forms with negligible cross-reactivity to liver or muscle PFK isoforms, enabling selective immunoblot and immunofluorescence visualization in mouse tissue homogenates and cultured cells.
