Target Information
Rat placental alkaline phosphatase (ALPP) is a 524-residue, glycosyl-phosphatidylinositol-anchored ectoenzyme whose extracellular domain contains two zinc ions and one magnesium ion coordinated within the characteristic D/SxDxGxGxGxxD motif, enabling selective hydrolysis of phosphate monoesters at alkaline pH. The protein is tethered to the apical brush border of syncytiotrophoblasts and released as a 140 kDa homodimer after GPI cleavage. Expressed almost exclusively in placenta and at low levels in uterus, ALPP localizes to microvilli and maternal-facing membranes. Antibodies raised against the catalytic or GPI-anchor proximal peptide detect native, glycosylated and secreted dimers with negligible cross-reactivity to intestinal or bone alkaline phosphatase isoforms, enabling selective immunoblot and immunohistochemical visualization in rat placental homogenates and serum.
