Target Information
Triggering receptor expressed on myeloid cells 1 (TREM1) is a cell surface receptor integral to enhancing both innate and adaptive immunity by amplifying inflammatory reactions. When activated by ligands such as PGLYRP1, HMGB1, or HSP70, TREM1 multimerizes and partners with the transmembrane adapter TYROBP/DAP12. This interaction triggers a SYK-mediated cascade of tyrosine phosphorylation, which in turn activates several downstream mediators, including BTK, MAPK1, MAPK3, and phospholipase C-gamma. In mice, TREM1 is involved in various functions such as inflammatory response, immune regulation, tumor progression, and neurodegenerative diseases. Mouse models show that the absence of TREM1 can mitigate the inflammatory response induced by lipopolysaccharide (LPS), decrease levels of proinflammatory cytokines, and enhance survival rates. Additionally, TREM1 plays a role in regulating bone marrow and granulocyte functions, with its loss leading to decreased granulocyte numbers and impaired function.
