Target Information

Sphingosine kinase 1 is responsible for converting sphingosine into sphingosine 1-phosphate (SPP), a lipid mediator with roles both inside and outside the cell. It can also phosphorylate D-erythro-sphingosine and, to a lesser extent, sphinganine, but does not act on other lipids like D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. Sphk1 is crucial for regulating glucose metabolism and insulin signaling pathways. In mouse models of type 2 diabetes, such as KK/Ay mice, Sphk1 overexpression markedly enhanced both basal and insulin-stimulated glucose uptake, improved insulin sensitivity, and lowered blood glucose levels. Furthermore, Sphk1 facilitated glycogen synthesis by activating Akt and inhibiting GSK3β, thus enhancing liver function and glucose metabolism.
