Target Information

Optineurin is crucial for maintaining the Golgi complex, facilitating membrane trafficking, and supporting exocytosis through its interactions with myosin VI and Rab8. It connects myosin VI to the Golgi complex and is vital for forming the Golgi ribbon. Optineurin is also involved in activating the innate immune response during viral infections. In mice, Optineurin has a broad range of functions, including roles in autophagy, immune response, muscle regeneration, neuroprotection, bone metabolism, and more. Mutations related to ALS, such as E478G, can impair Optineurin's function, resulting in the obstruction of mitochondrial autophagy. Additionally, Optineurin plays a part in Parkin-mediated selective mitochondrial autophagy (mitophagy), helping to maintain mitochondrial quality within cells.
