Target Information
The mitochondrial branched-chain-amino-acid aminotransferase catalyzes the initial reaction in the catabolism of the essential branched-chain amino acids leucine, isoleucine, and valine. It may also serve as a transporter for branched-chain alpha-keto acids. In mouse skeletal muscle, BCAT2 is a vital enzyme for the metabolism of branched-chain amino acids (BCAAs). It facilitates the transamination reaction of BCAAs, marking the first step in BCAA metabolism and playing a crucial role in maintaining amino acid balance within the body. In BCAT2 knockout mice, plasma levels of branched-chain amino acids increase, while levels of branched-chain keto acids (BCKAs) decrease, highlighting BCAT2's significant role in regulating amino acid metabolism. Additionally, these knockout mice exhibit higher energy expenditure and improved insulin sensitivity, along with enhanced glucose tolerance, which may be connected to the disruption in BCAA metabolism.
