Target Information

The cytotoxic effect of Bactericidal Permeability-Increasing Protein (BPI) is primarily targeted at various Gram-negative bacteria. This specificity is likely due to the strong affinity of its very basic N-terminal half for the negatively charged lipopolysaccharides unique to the outer envelope of Gram-negative bacteria. In mouse models, BPI demonstrates significant antibacterial, anti-endotoxin, and immunomodulatory properties, establishing it as a promising candidate for treating Gram-negative bacterial infections and sepsis. BPI not only directly kills bacteria but also neutralizes the lipopolysaccharides (LPS) released by these bacteria, thereby mitigating the inflammatory response induced by LPS. For instance, in a mouse sepsis model, BPI effectively lowers serum levels of tumor necrosis factor-α (TNF-α) and interleukin-1α (IL-1α), protecting mice from lethal endotoxin shock.
