Chicken Phospholamban (PLN) is a small integral membrane protein that plays a crucial role in regulating cardiac function by modulating the activity of the sarcoplasmic reticulum Ca²⁺-ATPase (SERCA). PLN inhibits SERCA in its unphosphorylated state, reducing calcium uptake into the sarcoplasmic reticulum and decreasing cardiac contractility. When PLN is phosphorylated by protein kinase A (PKA) or other kinases, its inhibitory effect on SERCA is relieved, leading to increased calcium uptake and enhanced cardiac relaxation. This regulatory mechanism is essential for maintaining proper calcium homeostasis and cardiac function. Abnormal expression or mutations in PLN can lead to cardiomyopathies and heart failure.
