Target Information

Mouse Bruton's tyrosine kinase (BTK) is a 76 kDa cytoplasmic kinase whose N-terminal PH-TH module, SH3-SH2 cassette, and C-terminal kinase lobe form a clamp that phosphorylates PLCγ2 and other B-cell signaling substrates after membrane recruitment. A solvent-exposed 16-aa "KVE" loop (aa 340-355) between the SH2 and kinase domains folds into a β-hairpin unique among mouse Tec kinases, presenting a conformational epitope preserved after autophosphorylation and fixation. Affinity-purified antibodies targeting this loop trace native BTK in splenic B-cell follicles, bone-marrow pre-B cells, and peritoneal macrophages, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to Tec or Itk.
