Target Information

Canine endothelial nitric-oxide synthase (eNOS, NOS3) is a 133 kDa homodimeric flavo-heme enzyme whose oxygenase domain converts L-arginine to nitric oxide while its reductase domain shuttles electrons from NADPH through FAD and FMN. A solvent-exposed 16-aa "KVE" loop (aa 495-510) between the heme and BH4 pockets folds into a β-hairpin unique among canine NOS isoforms, presenting a conformational epitope preserved after phosphorylation and fixation. Affinity-purified antibodies targeting this loop trace native eNOS in pulmonary artery endothelium, cardiac capillaries, and renal glomeruli, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to nNOS or iNOS.
