Tumor necrosis factor receptor superfamily member 5 serves as a receptor for TNFSF5/CD40LG. It facilitates TRAF6- and MAP3K8-mediated signaling pathways that activate ERK in macrophages and B cells, resulting in the stimulation of immunoglobulin secretion. CD40's roles in mice are vast and intricate, encompassing immune regulation, inflammatory responses, tumor immunity, autoimmune diseases, and organ damage. The interaction between CD40 and CD40L is essential for Th1 and Th2 cells to activate B cells. Mice deficient in CD40 struggle to effectively activate B cells. In the experimental autoimmune encephalitis (EAE) model, CD40 expression in the central nervous system significantly influences the disease's development and progression, with CD40-deficient mice exhibiting milder pathological changes and inflammatory responses.
