Target Information
Sestrin-1 acts as an intracellular sensor for leucine and plays a crucial role in negatively regulating the TORC1 signaling pathway via the GATOR complex. In the absence of leucine, Sestrin-1 binds to the GATOR2 subcomplex, inhibiting TORC1 signaling. However, when leucine binds to SESN2, this interaction with GATOR2 is disrupted, activating the TORC1 signaling pathway. In mice, Sestrin-1 is involved in various functions, including anti-oxidation, anti-aging, metabolic regulation, and protection of muscles, heart, brain, and immune system. It has demonstrated protective effects in mouse models, such as minimizing cell damage due to oxidative stress by inhibiting mTORC1.
