Defects in the Klotho gene can lead to various pathological phenotypes in mice that resemble human aging, such as arteriosclerosis, osteoporosis, emphysema, cognitive impairment, and a shortened lifespan. Conversely, overexpressing Klotho can extend lifespan and mitigate aging-related phenotypes in mice, such as cognitive decline and reduced muscle strength.
