Target Information
Klotho may possess weak glycosidase activity towards glucuronylated steroids, although it lacks crucial active site Glu residues at positions 241 and 874, indicating it might be inactive as a glycosidase in vivo. Klotho is thought to play a role in regulating calcium and phosphorus homeostasis by inhibiting the synthesis of active vitamin D. It is crucial for the specific interaction between FGF23 and FGFR1. In mice, Klotho is associated with a variety of functions, including anti-aging, metabolic regulation, cardiovascular protection, nervous system maintenance, immune regulation, and bone health. Defects in the Klotho gene can lead to various pathological phenotypes in mice that resemble human aging, such as arteriosclerosis, osteoporosis, emphysema, cognitive impairment, and a shortened lifespan. Conversely, overexpressing Klotho can extend lifespan and mitigate aging-related phenotypes in mice, such as cognitive decline and reduced muscle strength. Additionally, Klotho exerts its anti-aging effects by inhibiting the insulin and insulin-like growth factor 1 (IGF1) signaling pathways.
