Target Information

Interleukin-6 (IL-6) is a cytokine with diverse biological functions related to immunity, tissue regeneration, and metabolism. It binds to IL6R, forming a complex with the signaling subunit IL6ST/gp130 to activate the intracellular IL6-signaling pathway. IL-6 is a strong inducer of the acute phase response, playing a crucial role in host defense during infection and tissue injury. However, excessive IL-6 production is associated with disease pathology. In mice, IL-6 is involved in multiple physiological and pathological processes, including liver regeneration, immune response, muscle atrophy, obesity, and angiogenesis following cerebral ischemia. It is essential for liver regeneration, as studies have shown that IL-6 deficient mice experience significant liver regeneration impairment after liver resection, marked by hepatocyte necrosis and functional failure. Additionally, IL-6 deficiency results in decreased hepatocyte proliferation capacity and disrupted DNA synthesis signaling pathways, such as loss of STAT3 activation and reduced expression of AP-1, Myc, and Cyclin D1. These defects, however, can be remedied by supplementing IL-6.
