Target Information
Clusterin acts as an extracellular chaperone that prevents the aggregation of non-native proteins, helping to avert stress-induced aggregation of blood plasma proteins. It inhibits the formation of amyloid fibrils by proteins such as APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants. In mice, clusterin has a wide range of functions, including roles in apoptosis, neuroprotection, metabolic regulation, immune response, nephropathy, and retinal development. It is closely linked to metabolic diseases, with research showing that its overexpression can protect mice from obesity and non-alcoholic fatty liver disease (NAFLD) induced by a high-fat Western diet. This protective effect is attributed to clusterin's antioxidant and anti-inflammatory actions via the Nrf2 pathway.
