Target Information

Oligomeric Apaf-1 facilitates the cytochrome c-dependent autocatalytic activation of pro-caspase-9 (Apaf-3), which in turn activates caspase-3, leading to apoptosis. This activation process requires ATP. In mice, Apaf1 has diverse roles, including regulating apoptosis, suppressing immune responses, maintaining mitochondrial function, and supporting cell survival. Its functions extend beyond cell death, encompassing biological processes like immune regulation, stress response, and development. As a core component of the apoptosome, Apaf1 binds to cytochrome c to activate caspase-9, thereby initiating an apoptosis cascade, which is vital for cell death and embryonic development. For instance, mouse embryonic fibroblasts lacking Apaf1 exhibit abnormalities in mitochondrial morphology and function, impacting cytochrome c release and the basic metabolic state, highlighting Apaf1's critical role in regulating the mitochondrial apoptosis pathway.
