Rat programmed cell death protein 1 ligand 1 (PD-L1, CD274) is a 32 kDa type-I transmembrane glycoprotein whose IgV-like domain and short cytoplasmic tail engage PD-1, delivering inhibitory cues to T cells. A solvent-exposed 14-aa "KVE" loop (aa 135-148) between the CC' and F strands of the IgV fold folds into a β-hairpin unique among rat B7 family members, presenting a conformational epitope preserved after glycosylation and fixation. Affinity-purified antibodies targeting this loop trace native PD-L1 in splenic dendritic cells, alveolar epithelium, and cardiac endothelium, delivering crisp membrane rings and single-band clarity on immunoblots with no cross-reaction to PD-L2 or other B7 ligands.
