Target Information
Rat leukocyte elastase inhibitor (SERPINB1) is a 43 kDa intracellular serpin whose reactive-center loop (Phe-Leu-Met-Lys) docks into neutrophil elastase, forming a stable 1:1 complex that blocks proteolytic activity. A solvent-exposed 14-aa "KVE" loop (aa 340-353) between strand 5A and the hinge region folds into a β-hairpin unique among rat serpins, presenting a conformational epitope preserved after complexation and fixation. Antibodies targeting this loop trace native SERPINB1 in bone-marrow neutrophils, alveolar macrophages, and splenic granulocytes, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to α1-antitrypsin or other SERPIN family members.
