Target Information
Rat cytokeratin 14 (CK14, KRT14) is a 50 kDa type I intermediate filament protein comprising 472 residues. It pairs with keratin 5 to form stable heterodimers that assemble into 10 nm tonofilaments, anchored at hemidesmosomes via its N-terminal head domain and L12 linker. Expressed constitutively in basal keratinocytes of stratified epithelia, mammary myoepithelium and stratified squamous mucosa, CK14 localizes to cytoplasmic filament bundles and perinuclear cages. The protein undergoes phosphorylation during mitosis and binds plectin at residues 1-75. Antibodies raised against rod domain or C-terminal peptide detect native, phosphorylated and soluble forms with negligible cross-reactivity to other keratins, enabling precise immunoblot and immunohistochemical detection in rat skin, esophagus and cultured primary keratinocytes.
