Target Information
Rat cluster of differentiation 6 (CD6) is a 655-residue type I transmembrane glycoprotein whose extracellular region contains three scavenger-receptor cysteine-rich (SRCR) domains that bind CD166 (ALCAM). A single transmembrane helix anchors the protein to thymocytes and mature T cells, while a 44-aa cytoplasmic tail carries two tyrosine-based motifs recruiting SLP-76 and other adaptors. Constitutively expressed in spleen, thymus and lymph nodes, CD6 localizes to the immunological synapse and microvilli. Antibodies raised against SRCR3 or cytoplasmic peptide detect native, glycosylated and proteolytically shed forms with negligible cross-reactivity to CD5 or other SRCR proteins, enabling selective immunoblot and immunofluorescence analysis in rat lymphoid tissue homogenates and cultured immune cells.
