Target Information
Mouse octamer-binding transcription factor 1 (Oct-1, Pou2f1) is a 743-residue nuclear protein containing a bipartite POU domain: a 75-aa POU-specific box and a 60-aa POU homeobox that together recognize the consensus 5'-ATGCAAAT-3'. The N-terminal transactivation region is rich in serine, proline and glutamine, while a C-terminal leucine zipper mediates dimerization. Expressed ubiquitously in all tissues, Oct-1 localizes to euchromatin and heterochromatin, regulating housekeeping and cell-type-specific genes. Antibodies raised against the POU domain or N-terminal epitope detect native, phosphorylated and alternatively spliced isoforms with negligible cross-reactivity to Oct-2 or other POU factors, enabling selective immunoblot, immunohistochemistry and chromatin immunoprecipitation in mouse tissue homogenates and cultured cells.
