Target Information

Prostaglandin E2 (PGE2) serves multiple roles in mice, encompassing immunoregulation, inflammatory response, bone metabolism, tumor development, and neurobehavior. PGE2 modulates the immune response through its receptors, EP2 and EP4. The EP2 receptor primarily dampens the immune response by inhibiting T cell activation and proliferation. In terms of inflammatory response, PGE2 is particularly crucial in activating macrophages. Research indicates that PGE2 enhances the activation of NLRP3 inflammasomes in macrophages via TLR2 and TLR4 signaling pathways, thereby influencing the inflammatory response. Additionally, PGE2 exerts an immunosuppressive effect during infections by inhibiting NADPH oxidase activity and decreasing bacterial killing capability. PGE2 is a key driver of bone resorption, promoting this process by boosting osteoclast activity. However, when EP4 receptors are absent, PGE2's inhibitory effect on bone resorption is heightened, resulting in reduced bone resorption.
