Target Information

Inosine triphosphate pyrophosphatase is an enzyme that hydrolyzes non-canonical purine nucleotides such as inosine triphosphate (ITP), deoxyinosine triphosphate (dITP), 2'-deoxy-N-6-hydroxylaminopurine triphosphate (dHAPTP), and xanthosine 5'-triphosphate (XTP) into their corresponding monophosphate derivatives. In mice, the role of ITPase extends beyond regulating nucleotide metabolism; it also encompasses contributions to nervous system development, maintaining genome stability, and influencing susceptibility to certain diseases. A deficiency in ITPase can result in the depolarization of neurons and trigger epileptic seizures. For instance, mice with ITPase deficiency specifically in neural stem cells (Itpa-cKO) exhibit growth retardation, epileptic seizures, and developmental delays.
