Heat shock protein HSP 90-beta is a molecular chaperone that facilitates the maturation, structural maintenance, and regulation of specific target proteins involved in processes such as cell cycle control and signal transduction. Its activity is linked to a functional cycle driven by ATPase activity, which likely induces conformational changes in client proteins, activating them in the process. It dynamically interacts with various co-chaperones that influence its substrate recognition, ATPase cycle, and chaperone function. By engaging with different classes of client proteins, HSP 90-beta utilizes various co-chaperone proteins or complexes that act as adapters, allowing simultaneous interaction with the specific client and the central chaperone. In a diet-induced obesity mouse model, Hsp90ab1 is upregulated in skeletal muscle and plays a role in regulating insulin signaling by impacting glucose and fatty acid metabolism. Research has shown that knocking out Hsp90ab1 can improve insulin resistance, lower blood glucose levels, enhance the activity of the pyruvate dehydrogenase complex, and improve glucose tolerance.
