Endonuclease 8-like 3 is a DNA glycosylase that preferentially targets single-stranded DNA (ssDNA) or partially ssDNA structures like bubble and fork formations over double-stranded DNA (dsDNA). It plays a role in interstrand cross-link repair during replication stress by cleaving one of the N-glycosyl bonds in the interstrand cross-link, preventing double-strand breaks while creating an abasic site that translesion synthesis polymerases bypass. NEIL3 is involved in repairing oxidatively damaged DNA, such as hydroxylamine and guanidinamine lesions, via the base excision repair (BER) pathway. Research indicates that NEIL3 exhibits high repair efficiency on single-stranded DNA, particularly in neural stem cells in the brain and bone marrow, a function critical for maintaining genome stability. Additionally, mice deficient in NEIL3 show reduced neuronal regeneration following brain injury, underscoring its vital role in DNA damage repair.
