Target Information

Actins are highly conserved proteins essential for various cell motility processes and are universally expressed in all eukaryotic cells. Mutations in the Acta2 gene are a primary cause of familial thoracic aortic aneurysms and dissections. These mutations result in VSMC dysfunction, characterized by excessive proliferation of vascular smooth muscle and irregular elastic fibers. For instance, Acta2-deficient mice exhibit dilation of the aortic root due to disordered elastic fibers and reduced extracellular matrix remodeling capacity. Additionally, Acta2 mutations increase blood vessels' sensitivity to angiotensin II (AngII), thereby exacerbating vasodilation and inflammatory responses.
