Target Information
Mouse Thyroid Peroxidase (TPO) is a crucial enzyme in thyroid hormone synthesis, primarily expressed in the thyroid gland. TPO catalyzes the oxidation of iodide ions to iodine and the iodination of tyrosine residues in thyroglobulin, which are essential steps in the production of thyroid hormones, triiodothyronine (T3) and thyroxine (T4). TPO is located in the apical membrane of thyroid follicular cells and is regulated by thyroid-stimulating hormone (TSH). TPO is also involved in the pathogenesis of autoimmune thyroid diseases such as Hashimoto's thyroiditis and Graves' disease. In these conditions, autoantibodies against TPO (TPOAb) are produced, leading to thyroid dysfunction. TPOAb levels are used as diagnostic and prognostic markers for these diseases. In addition, TPO has been implicated in other thyroid disorders, including congenital hypothyroidism, where mutations in the TPO gene can result in impaired thyroid hormone synthesis. TPO is a potential therapeutic target for the treatment of thyroid diseases.

User Note
- Always centrifuge tubes before opening. Avoid mixing by vortexing or pipetting. Reconstitute in 10 mM PBS (pH7.4) to a concentration of 0.1-1.0 mg/mL. Aliquot the reconstituted solution to minimize freeze-thaw cycles.
