Target Information

Mouse Semaphorin-3E (Sema3e) is a secreted protein that plays a role in various biological processes, including axon guidance, immune regulation, and vascular development. It binds to its receptor PlexinD1 to regulate cell migration and proliferation. In cancer, Sema3e has been shown to inhibit tumor growth by suppressing angiogenesis and promoting apoptosis. In diabetic retinopathy, Sema3e is secreted by severely ischemic retinal neurons and activates PlexinD1 on endothelial cells to inhibit VEGF-induced angiogenesis, thereby normalizing revascularization and suppressing extra retinal vascular outgrowth. In allergic asthma, Sema3e expression is significantly suppressed in the airways of severe asthmatic patients, and genetic ablation of Sema3e in mice results in increased lung granulocytosis and airway hyper-responsiveness. Additionally, Sema3e has been associated with atherosclerosis, where it attenuates neointimal formation by suppressing vascular smooth muscle cell migration and proliferation. Its dysregulation is implicated in various diseases, making it a potential therapeutic target.
