Target Information
Mouse Phospholamban (Pln) is a small protein that plays a crucial role in regulating cardiac contractility and calcium cycling. It inhibits the sarcoplasmic reticulum Ca2+-ATPase (SERCA2a), which is responsible for calcium uptake and cardiac relaxation. Phosphorylation of Pln by protein kinase A (PKA) or Ca2+/calmodulin-dependent protein kinase II (CaMKII) relieves this inhibition, enhancing cardiac function. In mouse models, Pln deficiency leads to increased cardiac contractility and reduced responsiveness to β-adrenergic stimulation. Conversely, overexpression or mutations in Pln, such as the R14del variant, can cause cardiomyopathies and heart failure due to impaired SERCA2a function and calcium handling. These studies highlight Pln's essential role in maintaining cardiac function and its potential as a therapeutic target for heart diseases.
