Target Information
In mice, Pappalysin-1 (Pappa) is a zinc-binding metalloproteinase that plays critical roles in various physiological and pathological processes. Pappa cleaves insulin-like growth factor-binding proteins (IGFBP-4 and IGFBP-5), releasing insulin-like growth factors (IGF) to activate the IGF signaling pathway. This pathway is essential for cell growth, proliferation, and differentiation. Pappa is involved in skeletal development and bone formation. Studies show that Pappa knockout mice exhibit reduced bone mineral density, while overexpression of Pappa increases bone growth. Additionally, Pappa has been implicated in cancer progression, where it promotes tumor cell proliferation and invasion by enhancing IGF bioavailability and activating signaling pathways such as NF-κB and PI3K/AKT.

User Note
