Target Information
Mouse Optineurin (Optn) is a multifunctional polyubiquitin-binding adaptor protein involved in various cellular processes, including autophagy, vesicle trafficking, and immune signaling. It is highly expressed in the brain, spinal cord, spleen, heart, liver, lung, and skeletal muscles. Optineurin binds to methionine 1- and lysine 63-linked polyubiquitin chains, regulating processes like NF-κB signaling and type I interferon production. In immunity, Optineurin is crucial for defense against cytosolic bacteria via selective autophagy (xenophagy) and modulates cytokine secretion. Optineurin deficiency in mice leads to chronic microglial activation and increased susceptibility to bacterial infections. It also plays a role in neurodegenerative diseases such as Amyotrophic Lateral Sclerosis (ALS) and Glaucoma.
