Mouse Nephrin (Nphs1) is a critical transmembrane protein component of the slit diaphragm, a key structure in the glomerular filtration barrier. It is essential for maintaining the selective permeability of the glomerular filtration barrier, allowing small molecules to pass while retaining essential proteins and blood cells. In mouse models, Nphs1 is highly expressed in podocytes, where it forms a molecular sieve that contributes to the proper functioning of the glomerular basement membrane (GBM). Abnormal expression or dysfunction of Nphs1 can lead to various kidney diseases, including nephrotic syndrome and diabetic nephropathy. Additionally, Nphs1 has been implicated in the regulation of insulin release from pancreatic beta cells, highlighting its broader physiological roles.
