Rat HAS1 is a multi-pass integral membrane enzyme with conserved cytoplasmic catalytic domains facing the cell interior. It maintains stable transmembrane topology and intact sugar substrate binding motifs under physiological conditions, embedded within plasma membranes of connective tissue cells. Its membrane-embedded catalytic structure and dual-sided domain layout separate it from soluble cytoplasmic enzymes, secreted circulating glycoproteins and intranuclear transcription factors with simplified structural frameworks and fixed single-compartment localization patterns.
