Mouse hemicentin 1 (HMCN1) is a 5,635-residue extracellular scaffold built from 48 Ig-like modules, 21 EGF-like repeats (16 Ca²⁺-binding), two von Willebrand A domains and a unique 450-residue fibulin-like insert. The polypeptide forms disulfide-bonded trimers that integrate into basement membranes and elastic fiber interfaces. Expressed in skin, lung and vascular endothelium, it localizes to dermal-epidermal junctions and Bruch's membrane. Antibodies against the fibulin-like region or VWA1 detect native, glycosylated and proteolytic fragments with negligible cross-reactivity to fibulin-1 or-2, enabling selective immunoblot and immunohistochemistry in mouse tissue homogenates and sections.
