Target Information

Mouse diacylglycerol kinase gamma (DGKγ) is a 70 kDa membrane-associated kinase whose N-terminal EF-hands, C1 domains and C-terminal catalytic pocket convert DAG to phosphatidic acid, using ATP to terminate lipid signaling. A solvent-exposed 16-aa "KVE" loop (aa 420-435) between the kinase core and the MARCKS-homology region folds into a β-hairpin unique among mouse DGK isoforms, presenting a conformational epitope preserved after membrane binding and fixation. Affinity-purified antibodies targeting this loop trace native DGKγ in cortical synapses, hippocampal dendrites, and cardiac sarcolemma, delivering crisp punctate membranes and single-band clarity on immunoblots with no cross-reaction to DGKα or DGKζ.
