Target Information
Mouse 5T4 is a 434-residue type I transmembrane glycoprotein whose extracellular segment contains seven leucine-rich repeats flanked by juxtamembrane EGF-like and thyroglobulin-type modules; a single transmembrane helix anchors the protein to the plasma membrane with a short cytoplasmic tail lacking catalytic motifs. The molecule is heavily N-glycosylated, adopts a curved solenoid fold, and forms non-covalent dimers at the cell surface. Constitutively expressed on placental trophoblasts and developing epithelia, 5T4 localizes to filopodia and microvilli. Antibodies raised against the LRR-EGF junction or C-terminal peptide detect native, glycosylated and shed forms with negligible cross-reactivity to other LRR proteins, enabling selective immunoblot and immunohistochemical visualization in mouse tissue homogenates and cultured cells.
