Target Information
Mouse nischarin is a 1,657-residue scaffold protein built from an N-terminal LIM domain, five PDZ modules, a coiled-coil region and a C-terminal alpha-helical segment that collectively mediate binding to integrin α5, PAK1 and the exocyst complex. Predominantly cytosolic, the polypeptide localizes to focal adhesions, lamellipodia and early endosomes in epithelial and vascular smooth-muscle cells. Alternative splicing introduces a 33-residue insert between PDZ3 and PDZ4, generating tissue-specific isoforms of 1657 and 1624 amino acids. Antibodies raised against the unique linker or full-length recombinant nischarin recognize native, phosphorylated and splice-variant forms with negligible cross-reactivity to other LIM/PDz proteins, enabling precise immunoblot and immunofluorescence detection in mouse tissue homogenates and cultured cells.
