Target Information
Sortilin operates as a sorting receptor within the Golgi compartment and as a clearance receptor on the cell surface. It is essential for protein transport from the Golgi apparatus to the lysosomes via a pathway independent of the mannose-6-phosphate receptor (M6PR). In mice on a high cholesterol diet, sortilin deficiency leads to reduced hepatic bile acid synthesis without impacting cholesterol ester secretion. Additionally, sortilin deficiency decreases free cholesterol accumulation in the liver, thereby lowering the risk of gallstones. Widely expressed in the mouse brain, particularly in the prefrontal cortex and hippocampus, sortilin functions as a receptor for nerve growth factors, such as neurotrophin NTS3/Sortilin, and plays a role in neuronal survival, differentiation, and synaptic plasticity.
