Target Information

Granzyme A is a predominant protease found in the cytosolic granules of cytotoxic T-cells and NK-cells, known for inducing caspase-independent pyroptosis when delivered into target cells via the immunological synapse. It specifically cleaves after lysine (Lys) or arginine (Arg) residues. Granzyme A cleaves the APEX1 protein at 'Lys-31', impairing its oxidative repair functionality. It also cleaves the nucleosome assembly protein SET at 'Lys-189', disrupting its ability to assemble nucleosomes and enabling the SET complex to move into the nucleus to nick and degrade DNA. In mice, Granzyme A's functions are primarily associated with pro-inflammatory effects, antiviral and antibacterial immune responses, tumor suppression, and immune regulation. Its pro-inflammatory effects are particularly noteworthy in inflammatory diseases, and inhibiting Granzyme A could offer a novel strategy for treating certain conditions.
