Target Information

Lysosomal alpha-glucosidase is crucial for glycogen degradation within lysosomes. It exhibits the highest activity on alpha-1,4-linked glycosidic linkages but can also hydrolyze alpha-1,6-linked glucans. In mice, the function of lysosomal acid alpha-glucosidase (GAA) is closely associated with hereditary glycogen storage disease type II (GSDII or Pompe disease). GAA is the lysosomal enzyme responsible for breaking down α-1,4 and α-1,6 bonds in glycogen. A deficiency in GAA results in the accumulation of glycogen within lysosomes, leading to cell and tissue damage. Mice with a knockout of the GAA gene exhibit significant glycogen accumulation in the heart and skeletal muscles, accompanied by abnormal dilation and vacuolization of lysosomes.
