G6PD is the initial enzyme in the pentose phosphate pathway (PPP), primarily responsible for catalyzing the conversion of glucose-6-phosphate into 6-phosphogluconolactone, while producing NADPH. NADPH serves as a vital reducer within cells, engaging in antioxidant processes and safeguarding cells from oxidative stress. G6PD is an X-linked "housekeeping" enzyme ubiquitously expressed in all mammalian cells. In mice, the G6PD gene resides on the X chromosome, with its activity being influenced by sex and X chromosome inactivation. For instance, in female mouse oocytes, G6PD synthesis relies on X chromosome activity. The expression and function of G6PD differ across tissues. In the mouse liver, G6PD's synthesis and degradation rates remain stable regardless of inducement or inhibition, indicating a level of consistent expression. Moreover, localization studies in mouse kidneys reveal that G6PD predominantly resides in peroxisomes.
