Target Information
Crystallins are the primary structural components of the vertebrate eye lens. In mouse retinal astrocytes, βA1-crystallin mainly functions to regulate glucose metabolism and mitochondrial function. This regulation is achieved by modulating the activity of protein tyrosine phosphatase 1B (PTP1B). βA1-crystallin binds to PTP1B and inhibits its phosphatase activity through an allosteric site, rather than the active site, ensuring normal glucose metabolism. Under conditions of hyperglycemic stress, the absence of βA1-crystallin results in increased PTP1B activity, which in turn inhibits the STAT3 signaling pathway. This leads to disturbances in glucose metabolism, heightened oxidative stress, and an inflammatory response.
