Target Information

Asporin binds to calcium and is involved in osteoblast-mediated collagen biomineralization. It serves as a key regulator of TGF-beta in articular cartilage, playing a vital role in maintaining cartilage homeostasis and contributing to the pathogenesis of osteoarthritis (OA). Asporin inhibits chondrogenesis in the articular cartilage by preventing the interaction between TGF-beta and its receptors on the cell surface, thus obstructing the canonical TGF-beta/Smad signaling pathway. In mice, the absence of asporin results in skin thickening and significant alterations in the content and structure of glycosaminoglycans (GAGs) within the skin. In periodontal tissues, asporin negatively impacts osteoblast differentiation and mineralization by suppressing the TGF-β1 signaling pathway, leading to changes in the structure of the periodontal ligament and enhanced bone resorption in mice lacking asporin. Furthermore, in articular cartilage, asporin helps maintain the differentiation of chondrocytes by inhibiting the TGF-β1 signaling pathway, thereby protecting against cartilage degradation.
