Interleukin-12 (IL-12) is a heterodimeric cytokine crucial for cell-mediated immune responses. IL-12 is a primary driver of Th1 cell differentiation, which is essential for cell-mediated immunity. Th1 cells produce interferon-gamma (IFN-γ), a key cytokine for defense against intracellular pathogens. The IL-12p40 subunit is a part of the active cytokines interleukin-12 and interleukin-23. Expressed by activated macrophages, this cytokine is pivotal in inducing the development of Th1 cells. Guinea pigs are frequently used to study DTH reactions, which are Th1-mediated immune responses. IL-12 plays a critical role in these reactions.
