Target Information

Nicotinamide N-methyltransferase (NNMT) facilitates the N-methylation of nicotinamide by utilizing the universal methyl donor S-adenosyl-L-methionine to produce N1-methylnicotinamide and S-adenosyl-L-homocysteine, serving as a primary pathway for nicotinamide/vitamin B3 clearance. NNMT plays a crucial role in regulating cellular methylation potential by consuming S-adenosyl-L-methionine, thereby limiting its availability for other methyltransferases. In mice, NNMT is highly expressed in adipose tissue and liver, with its expression level strongly linked to obesity, insulin resistance, and metabolic syndrome. For instance, NNMT knockout mice on a high-fat diet exhibit weight loss, improved insulin sensitivity, and better blood sugar control.
