Target Information

Rat orexin A is a 33-amino-acid, C-terminally amidated neuropeptide excised from the 130-residue prepro-orexin precursor by prohormone convertases. The peptide contains four cysteines that form two intramolecular disulfide bridges (Cys6-Cys12, Cys7-Cys14), generating a compact cyclic structure essential for receptor binding. Expressed selectively in lateral and posterior hypothalamic neurons, orexin A is packaged in dense-core vesicles and released along axonal projections to thalamic and brainstem targets. Antibodies raised against the full cyclic peptide or C-terminal fragments detect native orexin A with negligible cross-reactivity to orexin B or other hypothalamic peptides, enabling precise immunohistochemical and immunoblot visualization in rat brain extracts and tissue sections.
