Target Information

Rat complement component 2 (C2) is a 734-residue serum glycoprotein synthesized primarily in liver and secreted as a 102 kDa single-chain proprotein. Its N-terminal CCP1-3 modules bind C4b, while a C-terminal trypsin-like serine-protease domain (His-Asp-Ser triad) remains latent until cleavage by C1s into 35 kDa C2b and 70 kDa C2a fragments. The activated C2a serine protease forms the classical C3 convertase C4b2a. Constitutively expressed and circulating at micromolar levels, C2 localizes to immune complexes and cell membranes upon activation. Antibodies raised against the C2a protease domain or full-length protein detect native, cleaved and complex-bound forms with negligible cross-reactivity to factor B or C1s, enabling selective immunoblot and immunohistochemical visualization in rat plasma and tissue sections.
