Mouse membrane protein, palmitoylated 6 (MPP6) is a palmitoylated transmembrane scaffold protein of the MAGUK family, a core regulator of epithelial cell polarity and membrane protein complex assembly. It features a conserved N-terminal palmitoylation motif for membrane anchoring, a central PDZ domain for protein-protein interaction, and a C-terminal SH3 domain critical for structural stability. It localizes predominantly to the basolateral membrane of mouse epithelial and neuronal cells, with negligible expression in mesenchymal and hematopoietic cells. The protein mediates scaffolding of membrane receptors and cytoskeletal components, with conserved motifs for modulating cellular junction integrity. Mouse MPP6 shares core functional domains with mammalian orthologs, and has distinct tissue specificity differentiating it from MPP1 and MPP3.
