Target Information
Mouse ATP-binding cassette transporter B11 (Abcb11), also termed bile salt export pump (BSEP), is a 1,604-residue polytopic ABC transporter of the ABCB subfamily. Its architecture comprises two membrane-spanning domains with six α-helices each, plus two cytosolic nucleotide-binding domains that hydrolyze ATP to drive conformational cycling. Predominantly localized to the canalicular membrane of hepatocytes, the pump translocates monovalent bile acids against steep concentration gradients. Expression is restricted to liver, with peak levels aligning with bile acid synthesis. Antibodies raised against cytoplasmic or external epitopes of mouse Abcb11 detect the native glycoprotein with negligible cross-reactivity to Abcb1 or Abcb4, enabling selective visualization in liver sections and isolated canalicular membranes.
