Target Information
Mouse alpha-hemoglobin-stabilizing protein (AHSP) is a 102-residue, 12 kDa cytosolic chaperone expressed exclusively in erythroid precursors. Its central β-barrel (residues 20-90) presents a hydrophobic groove that clamps free α-globin, preventing precipitation until assembly with β-globin. A conserved Tyr at position 52 coordinates heme iron, stabilizing the bound cofactor. The protein localizes to cytosol and ribosomal periphery during reticulocyte maturation. Antibodies raised against full-length recombinant mouse AHSP detect native, heme-bound and oxidized forms with negligible cross-reactivity to β-globin or other erythroid chaperones, enabling precise immunoblot and immunohistochemical detection in mouse hematopoietic tissues and cultured erythroid precursors.
